Tendulkar missed Two test matches against South Africa last month after the groin injury flared up.
He then took an extended rest before returning to action in a domestic Twenty20 tournament.
India will face Bangladesh and Pakistan in the limited-overs tri-series, scheduled to begin on 8 June.
As well as India and hosts Pakistan, the Asia Cup features Sri Lanka, Bangladesh, Hong Kong and the United Arab Emirates.
The tournament takes place between 24 June and 6 July.
Niranjan Shah, secretary of the Board of Control for Cricket in India, confirmed: "Tendulkar is not available. He has not recovered sufficiently."
